There is not enough permissions granted for the specified folder to the ASP.NET worker process account.
Please review the Windows Event Log (Application). There should be some Warnings related to this issue originating from ASP.NET; these warnings could contain exact information on what access has been requested by which account. This information will help you to assign the appropriate permissions.
