Would changing root password have any effects on how Plesk operates?

NO, and it is a good practice to change the root password often. Search these forums for security and locking down a plesk box. Disallow direct login access to root via shell, change your shell access from telnet (disable) to SSH2 and then make sure you only su - to root. Lots of things to do to secure your server from hackers. This is only a start.
