Question changing virtual host directory

Hello

My virtual directory path currently is on "C" drive and I want to move it to "D" drive .

The problem is that size of vhost directory is about 380 GB and I want a secure way to change the vhost directory.
the size is huge , It causes my websites to be down for a long time.

can I copy the files to the other drive and then just go to gui to change directory in a fast mode ?
think gui is not reliable in these operations , can you provide me a CLI way to do such ?
