How can change magic_quotes_gpc to ON in Plesk 8.2.1 and Zend Optimizer Installed?

Tried using a vhost.conf to set the PHP parameters?

Where is this located?

inside the conf directory. You'll need to SSH in as root to access it.

If you want to change magic_quotes_gpc globally you can change this in /etc/php.ini and reload apache afterwards.
